Targeting lactate dehydrogenase a improves radiotherapy efficacy in non-small cell lung cancer: from bedside to bench

Abstract Background Lactate dehydrogenase A ( LDHA) is overexpressed and associated with poor prognosis in many kinds of cancer. In the current study, we evaluated prognostic value LDHA expression non-small cell lung cancer (NSCLC), tested whether inhibition might improve radiotherapy efficacy NSCLC. Methods was investigated NSCLC patients, using online database further verified by immunohistochemistry. The Kaplan–Meier plotter database. vitro, two lines were pretreated oxamate, an inhibitor LDHA, colony formation method performed to determine cellular radiosensitivity. Comet assay used detect DNA damage after irradiation. Flow cytometry applied test cycle progression apoptosis, monodansylcadaverine (MDC) staining examine autophagy. Results Both mRNA protein levels up-regulated tissues. High a factor radioresistance patients. oxamate remarkably increased radiosensitivity both A549 H1975 cells, enhanced ionizing radiation (IR)-induced apoptosis autophagy, accompanied distribution alternations. Furthermore, induced reactive oxygen species (ROS) accumulation ATP depletion, which increase injury hinder repair activity. Conclusions Our study suggests that may be potential strategy needs clinical trials.
